4. LC50, EC50, and LD50 Calculation

  • LC50: The concentration of a substance that is lethal to 50% of a test population, typically aquatic organisms, within a specified time.

  • EC50: The concentration of a substance that causes a specific effect in 50% of a test population, often related to non-lethal responses like immobility and growth inhibition.

  • LD50: The dose of a substance that causes death in 50% of a test population, usually expressed as mg/kg body weight.

  • Calculate LC50, EC50, LD50 with 95% confidence intervals for acute and chronic toxicity tests.
  • The optimized analysis method is suggested based on the characteristics of the input data.

5. IC25 and IC50 Analysis

  • ICp is the inhibiting concentration for a (specified) percent effect. It represents a point estimate of a concentration of test material that is estimated to cause a designated percent impairment in a quantitative biological function, such as the size attained by organisms during a growth period.

  • For example, IC25 is the concentration estimated to result in organisms having a dry weight 25% lower than that attained by control organisms.

  • Estimate IC25 and IC50 with 95% confidence intervals for ecotoxicity and cytotoxicity tests by automatically selecting the best-fit model from five linear and non-linear regression models based on your data.
  • minimum of three replicates is necessary in a toxicity test to enable the calculation of the IC25 and IC50.

7. Estimation of NOEC and LOEC for a Single Observation

  • NOEC (No Observed Effect Concentration): The highest concentration of a test substance at which no statistically significant adverse effect is observed on the test organisms compared to the control group under defined experimental conditions.
  • LOEC (Lowest Observed Effect Concentration): The lowest concentration of a test substance at which a statistically significant adverse effect is observed on the test organisms compared to the control group under defined experimental conditions.
  • Comprehensive toxicity data analysis for a single endpoint (For Example: OECD Test Guideline No. 209: Activated Sludge, Respiration Inhibition Test (Carbon and Ammonium Oxidation), to determine the NOEC and LOEC for both acute and chronic toxicity studies.

  • The method for calculating NOEC and LOEC is referred to as Hypothesis Testing.

  • minimum of three replicates is necessary in a toxicity test to enable the calculation of the NOEC and LOEC.
  • To understand why a minimum of three replicates is required for toxicity test data to calculate NOEC and LOEC in ecotoxicity testing, and to explore detailed analysis methods, visit my blog.
